The organizer who invited me to speak at one of their events closed the business recently, and their website is no longer accessible, rendering the reference link I put in the application that I’m about to submit broken.
Is it a concern for Tech Nation? Should I try to change it with another evidence?

You can try to get a previous snapshot of the site from https://archive.org/
You can still try using the invite, traction and promotion posts from social platforms. Or try and get a reference letter from the organizer itself.
Screenshot, email invitation, programme event and reference from organiser should be fine. However , if not much is found about the speaking event, Some assessor may also suggest that it is not a sector leading event.
